A. Minimal Liquid Media (Broth)prefix = "o" ns = "urn:schemas-microsoft-com:office:office"

1. Dissolve a package media powder of a Minimal SD Base Medium and the appropriate dropout supplement to 0.5 L deionized H2O.

2. Adjust media pH to 5.8.

3. Autoclave at 121oC for 15min.

4. Store medium at room temperature.

 

B. Minimal Plating Media with Agar

1. Dissolve a package media powder of a Minimal SD Base Medium and the appropriate dropout supplement to 0.5 L deionized H2O.

2. Adjust media pH to 5.8.

3. Then add 2% agar, the agar will not dissolve until it is autoclaved.

4. Autoclave at 121oC for 15min. Cool to the temperature which your hand can endure it and then pouring plates.

5. Storage plated medium at room temperature. Store plates inverted at 4 oC.

6. Important: Verify that the agar you used contain no other nutriment!
